If you wind it a few turns and it doesn't start up, you can give it a little shake to get it started, whether it's an automatic or a manual wind. The intention is not to spin the rotor to impart more engergy to the mainspring, but simply to "unstick" the escapement from its locked position. The proper shake for this purpose is a quick turn back and forth, as if you were trying to make the hands spin.

Each escapement is set up slightly differently and the set-up and the condition of the lubricants will both effect how easily/quickly it will start up when wound. Even if takes a few more turns of the crown before it starts up, as long as it runs for a reasonable portion of its stated power reserve (say, 90% or so for a new-ish watch) and keeps good time, I wouldn't worry about it.
